Peptone Unveils Once-Monthly Obesity Drug Data... 30% Weight Loss Seen in Animal Study

Peptone has released the results of its preclinical and safety studies on 'PT403', a semaglutide-based obesity and diabetes treatment candidate administered once a month, at the American Diabetes Association (ADA) conference.
On June 8, Peptone announced that it had presented research findings related to PT403 at the American Diabetes Association (ADA 2026) annual scientific meeting held in New Orleans, United States.
PT403 is a long-acting injectable administered once a month, combining semaglutide— a GLP-1 receptor agonist (GLP-1RA)—with Peptone’s drug delivery platform, SmartDepot.
According to the company, in a high-fat diet-induced obese mouse model, the groups that received PT403 every two weeks or every three weeks showed approximately 30% weight loss at four weeks. In contrast, the groups that received semaglutide daily or every three days, used as comparators, did not achieve the same level of weight reduction.
The results of safety and tolerability assessments were also disclosed. In a study involving 16 healthy adults, those who received a single dose of PT403 showed no vomiting or nausea symptoms compared to the group that received weekly semaglutide. The main adverse events were mild injection site reactions and decreased appetite, and the company explained that the incidence of adverse drug reactions requiring therapeutic intervention was also low.
These research findings are considered data supporting the potential for developing long-acting GLP-1 formulations. Currently, GLP-1 class treatments used for obesity and diabetes are limited mainly by the need for repeated injections and gastrointestinal side effects. PT403 is being developed with the goal of maintaining sustained therapeutic effects with just a once-monthly injection.
